The following contains major spoilers for "Prelude to the New Dimension," from the pages of Giant-Size Amazing Spider-Man #1, on sale now from Marvel Comics.
One of the Multiverse's most surprising symbiote superheroes is officially making their way to the world of Earth-616, and no one saw it coming.
"Prelude to the New Dimension" (by Mitsuyasu Sakai, Gerardo Sandoval, Israel Silva, and VC's Joe Sabino, from the pages of Giant-Size Amazing Spider-Man #1) opens to find the members of the interdimensional Spider-Society trying to figure out exactly what to do about their latest unlikely ally, Kid Venom. After rescuing the young Ben Parker of Earth-982, Kid Venom is more than deserving of some help of his own, yet returning him to his own time isn't nearly as exact a science as returning him to his own universe. Unfortunately, the Spider-Society has no time to make any decision before Kid Venom's Clinter symbiote hears the call of Knull, and that is all it takes to send him rushing through a portal to the primary Marvel Universe of Earth-616 in search of his dark god.

Introduced in Taigami's "Kid Venom, Ep. 1" (from the pages of 2023's Death of the Venomverse #2), the young Kintaro, aka Sakata Kintoki, aka Kid Venom, was born in the late 900s in the Heian Period of Japanese history. In Kintaro's first appearance, he used his Clinter symbiote's powers to fight back a symbiote-possessed bear that had been ravaging a small fishing village. Not only did Kid Venom prove capable of pushing back against the threat at hand, but he also showcased his symbiote's ability to consume other symbiotes, eradicating them altogether.
More recently, in the pages of 2024's Kid Venom by Taigami and Mattia Iacono, the titular hero faced off against the members of the Symbiote Hive who had been running rampant and terrorizing his world. Eventually, this led Kintaro to become the unwitting protector of the young Benjamin Richard Parker of Earth-982, who had been pulled from his native reality to Kid Venom's by the Symbiote Hive. In keeping Ben safe, Kid Venom was able to make a stellar first impression on the heroes of the Spider-Society, whom he could very well need the help of going forward.

This Spider-Man Story Finally Answers the Mystery of the MCU's Earth-616
Marvel has just given fans the most succinct explanation for why the primary Marvel Universe is designated Earth-616, and it makes perfect sense.
The ancient, evil god known as Knull made his first full appearance in the pages of 2018's Venom #3 by Donny Cates and Ryan Stegman. Countless eons ago, Knull was content to reside in darkness. However, when the light of Celestial creation broke through that void, Knull struck out against these unwelcome interlopers by reaching into his own shadow and pulling out All-Black, the very first symbiote. While Knull was eventually imprisoned by the very symbiotes he created, he would return to terrorize the Marvel Universe all over again during the events of King in Black. Fortunately, Eddie Brock was ultimately able to slay Knull and claim his mantle as the King in Black, though that death clearly didn't take in its entirety for the progenitor of all symbiotes.
